Description
The potato was modified for resistance to Potato Virus Y infection. The modified potato expresses the viral coat protein to cross-protect the plant from viral inoculation, spread, multiplication and symptom expression. Although the mechanism is not well-defined, likely the production of the coat protein enhances the production of movement proteins and coordinates the formation of viral replication complexes, thus limiting the spread of the viral via cell-to-cell infections. Additionally, the modified potato expresses neomycin phosphotransferase II for kanamycin selection during transformation.

The modified potato (
Solanum tuberosum
) contains two genetic cassettes for expression of
Escherichia coli
neomycin phosphotransferase II (
nptII
) and Potato Virus Y (PVY) coat protein.
Transcription of
nptII
begins at an
Agrobacterium tumefaciens
nopaline synthase (
nos
) promoter and terminates at an
A. tumefaciens nos
terminator. Neomycin phosphotransferase is a selectible marker used for selecting transformants.
Transcription of PVY coat protein initiates from a Cauliflower Mosaic Virus 35S promoter and terminates at a
A. tumefaciens nos
terminator. The sequence of the PVY coat protein contains the last 17 codons of the viral replicase gene, the full coat protein sequence and the 3' noncoding region.

Western blot analysis indicated that NPTII was present and readily detected from leaf and tuber samples. From leaf extracts, the PVY coat protein was difficult to detect.
PCR analysis confirmed the presence of both
nptII
and PVY coat protein.
Southern blot analysis indicated three insertions of the PVY coat protein.
Northern blot analysis detected low levels of PVY coat protein transcript in root, stem and leaf tissues. Detection relied on extended autoradiographic exposure.
